Chloroquine phosphate, which has been used for more than 70 years and is on the World Health Organization’s List of Essential Medicines, the safest and most effective medicines needed in a health system. Chloroquine was discovered in 1934 by Hans Andersag but was initially ignored for a decade because it was considered too toxic for human use.

Chloroquine is a medication used to prevent and to treat malaria in areas where malaria is known to be sensitive to its effects. Certain types of malaria, resistant strains, and complicated cases typically require different or additional medication. Occasionally it is used for amebiasis that is occurring outside the intestines, rheumatoid arthritis, and lupus erythematosus. Does chloroquine and proguanil protect against malaria? Although chloroquine is an antimalarial, it is often not sufficient for prophylaxis. Chloroquine used to be the antimalarial of choice and it was widely used for from 1947 onwards. As a result, the malaria parasites endemic to certain regions developed a resistance to chloroquine. Chloroquine was an essential element of mass drug administration campaigns to combat malaria throughout the second half of the 20th century, and remains one of the World Health Organization’s essential medicines. However, after the malaria parasites Plasmodium falciparum and Plasmodium vivax began exhibiting resistance to the drug in the 1960s and 1980s, respectively, it was replaced by similar antimalarial compounds and combination therapies.

Aside from preventing and treating malaria, chloroquine is also occasionally used for amebiasis that is occurring outside the intestines, rheumatoid arthritis, and lupus erythematosus. During World War II, United States government-sponsored clinical trials for antimalarial drug development showed unequivocally that chloroquine has a significant therapeutic value as an antimalarial drug.
